How to Think About It
在于 treats the subject as a question of where the essence resides. 'X 的意义在于 Y' = 'X's significance lies in Y.' The 不...而... wrapper says 'not located at X, but located at Y' — geographical metaphor for where the truth sits. The 而 is the literary pivot 'rather' that gives the sentence its essay-like weight. This is not a phrase you'd use to settle a dinner argument; it's for sounding considered.

Common Mistake
Learners drop the 而 and write '不在于 X，在于 Y.' That's grammatical but reads as a list, not a redirection. The 而 is what signals 'rather/instead,' and without it you lose the rhetorical pivot.

Don't Confuse With
不是...而是...
不是...而是... is the everyday spoken version ('it's not X, it's Y'). 不在于...而在于... is reserved for talking about where the essence / point / meaning lies.

关键在于 = 'the key lies in' — single-sided. Use it when you only need to name the real point, not contrast it with a wrong one.
重要的不是...，而是...
Same redirection move but more colloquial. 不在于 sounds more analytic and abstract; 重要的不是 sounds more practical.
